Rapid Diagnosis of Bacteriuria by Measuring Bacterial Adenosine Triphosphate Using a Bioluminescence Assay
نویسندگان
چکیده
A rapid method for quantifying bacteriuria was evaluated in 114 clinical specimens using a bacterial adenosine triphosphate (ATP) assay. The procedure allows for removal and destruction of non-bacterial ATP and subsequent analysis of bacteria ATP by firefly (Photinus pyralis) luciferin -luciferase bioluminescence and requires 30 minutes as a whole. The correlation coefficient between the number of bacteria in urine samples and the ATP concentration was 0.889. Sensitivity and specificity representing more than 10,000 CFU/ml of bacteriuria were 90.0% and 96 .8%, respectively. Those representing more than 100,000 CFU/ml were 88.1% and 97.2%.

An Adenosine Triphosphate Bioluminescence Method for Evaluating the Microbial Contamination of the Salad-Preparing Tables and Salad-Serving Dishes in Restaurants of Mashhad City, Iran
Introduction: Consumption of vegetable products is increasing commonly in the world because they are recognized as an important source of nutrients, vitamins, and fiber for humans. Salads are among the most widely used foods that are also known as the most contaminated foods in restaurants. This study was conducted to determine the microbial contamination of salad-preparing tables and salad-ser...
متن کاملAnalysis of the disagreement between automated bioluminescence-based and culture methods for detecting significant bacteriuria, with proposals for standardizing evaluations of bacteriuria detection methods.
A fully automated method for detecting significant bacteriuria is described which uses firefly luciferin and luciferase to detect bacterial ATP in urine. The automated method was calibrated and evaluated, using 308 urine specimens, against two reference culture methods. We obtained a specificity of 0.79 and sensitivity of 0.75 using a quantitative pour plate reference test and a specificity of ...

Background: The mitochondria are an important source of adenosine triphosphate (ATP) production in pre-implantation embryo. Therefore, the objective of this study was to investigate the effect of vitrification and in vitro culture of mouse embryos on their mitochondrial distribution and ATP content. Methods: The embryos at 2-PN, 4-cell and blastocyst stages were collected from the oviduct of st...

متن کاملAtp Bioluminescence Assay for Rapid Antibacterial Susceptibility Testing
We studied adenosine triphosphate (ATP) synthesized by bacteria in tube dilution cultures to devise a rapid antibacterial susceptibility test. The ATP level was determined with a firefly (Photinus pyralis) luciferin/luciferase bioluminescence assay, which allows a direct quantifying of bacteria above 10,000 CFU/ml. At sub-MIC, total bacterial ATP levels were paralleled by increased viable bacte...
